DescriptionCadherin-1 (not to be confused with the APC C activating protein CDH1), also known as CAM 120 80 or E-cadherin or uvomorulin, is a human protein encoded by the CDH1 gene. CDH1 has also been designated as CD324 (Differentiation Cluster 324). It is a tumor suppressor gene.

E-cadherin (epithelium) is the most thoroughly studied member of the cadherin family. It consists of 5 cadherin repeats in the extracellular domain (EC1 ~ EC5), a transmembrane domain and an intracellular domain that binds to p120-catenin and β-catenin. The intracellular domain contains a highly phosphorylated region, which is essential for β-catenin binding and therefore for E-cadherin function. [Citation needed] β-catenin can also be combined with α-catenin. Alpha-catenin is involved in the regulation of actin-containing cytoskeletal filaments. In epithelial cells, E-cadherin-containing intercellular junctions are usually adjacent to actin-containing filaments in the cytoskeleton.

FunctionCadherins are calcium-dependent cell adhesion proteins. They preferentially interact with themselves in a homophilic manner in connecting cells; cadherins may thus contribute to the sorting of heterogeneous cell types. CDH1 is involved in mechanisms regulating cell-cell adhesions, mobility and proliferation of epithelial cells. Has a potent invasive suppressor role. It is a ligand for integrin alpha-E/beta-7.
