Small Exosomal miRNA, Big Roles

Exosomes are lipid bilayer vesicles having 30-100 nm in diameter naturally released from most cell types by cell membrane inward budding. Consequently, exosome carries many parental cell components, such as functional proteins, lipids, and cellular metabolites. Additionally, abundant and various nucleic acids have also been identified in exosomes, including both coding and non-coding RNAs.

Among identified exosomal RNAs, microRNAs (miRNAs) are those small non-coding RNAs with about 22 nucleotides. The non-coding miRNAs are important regulators in post-transcriptional gene silencing through binding to complementary binding to the site of the 3’-untranslated region or open reading frame of mRNA. These exosomal miRNAs could be transferred between cells with exosome vesicles, and thus negatively regulating the biofunctions of recipient cells. The importance of miRNA is mainly embodied in its regulation of tumor cell activities, such as cell proliferation, survival, migration, invasion, and so forth. The exosomal miRNAs could serve as both suppressors and promoters for tumor cells, even also have an indirect influence on tumor microenvironment and immune system, emerging as promising biomarkers for minimally invasive diagnosis of tumors.

Exosomal miRNA Microarray

Currently, exosomal miRNAs are mainly profiled and analyzed by RNA sequencing and microarray technologies. Exosomal miRNA sequencing is directly sequencing the exosomal miRNA-based cDNA library by advanced next-generation sequencers, while, exosomal miRNA microarray captures and detects the target exosomal cDNA sequences through predesigned labeled nucleic acid probes on the chip. In essence, exosomal miRNA sequencing is to directly profile unknown miRNAs, and exosomal miRNA microarray is indirect profiling unknown miRNAs by known nucleic acid probes. Compared with exosomal miRNA sequencing, exosomal miRNA microarray has features (both advantages and limitations) of:

  • Easy to perform and relatively lower cost.
  • A large number of samples can be analyzed simultaneously.
  • No need for extensive and complex bioinformatic analysis.
  • Characterize the miRNA levels based on visualized fluorescence intensity.
  • Predesigned probes are required.
  • Not applicable to the identification of isoforms and novel genes.
