Current targets with B-cell depletion as the main mechanism include CD20 and the BlyS/APRIL system, of which CD20 is a transmembrane protein expressed on the surface of B cells, and targeting CD20 can distinguish precursor B cells, progenitor B cells, and plasma cells, remove autoimmune memory B cells, and reconstitute the transitional B cell population. The BlyS/APRIL system is an important target in systemic lupus erythematosus (SLE). BlyS is essential in the maturation of B cells. Also, BlyS is able to promote the activation, proliferation, and differentiation of T cells. In contrast, APRIL binds mainly to antigen-presenting B cells, binds to BCMA, and promotes long-term plasma cell survival. Related studies have shown that anti-BlyS/APRIL antibodies are potent agents to treat SLE.
