To find cell biomarkers throughout the past few decades, conventional cell culture procedures including two-dimensional (2D) cell culture and removing disease cells from their natural milieu have been the primary approaches. The most significant shortcoming of these approaches is their inability to discover clinically meaningful indicators and adequately depict the microenvironment. Besides, other main differences between 2D and 3D cell cultures have been illustrated in Figure 1.

In recent years, 3D cell culture methods have shown great promise for producing realistic tissue and disease models. Several studies have compared the various growth techniques and found that cancer cells grown in 3D models are more phenotypically stable and closer to the phenotype of the parent cell line than 2D-cultured cancer cells. Moreover, based on drug discovery-associated trials, drug resistance in cancer cells produced in 3D models is comparable with in vivo resistance. All these findings support the notion that 3D culture platforms can be utilized to identify more clinically relevant and disease characteristics-associated biomarkers.
