Researchers developed a function-based HTS method to rapidly identify functional antibodies (antagonists and agonists) targeting G protein-coupled receptors (GPCRs). This approach combined GPI-anchored antibody cell display with ligand-induced recruitment cell sorting and screening, connecting antibody genotype to phenotype and applying it to all GPCR targets. They validated the approach by recognizing an agonist and a set of antibody antagonists against the human apelin receptor from an immune repertoire. In contrast, phage display only yielded neutral binders and antagonists. This novel method demonstrates greater efficiency compared to traditional approaches, potentially setting a new standard in antibody drug discovery.
