Background

Mesothelin (MSLN) is a 303 aa GPI-anchored protein with a molecule weight of 40 kDa. MSLN creates soluble mesothelin-related peptides (SMRP), which have been employed as markers to detect malignant mesotheliomas. Mesothelin participates in multiple signaling pathways via interactions with soluble MSLN and CA125/MUC16 or by overexpression, to activate Akt, ERK 1/2, JNK, SGK3/FOXO3, p38, NF-κB, and STAT3 pathways, thus regulating the downstream effects like inhibition of Bim, Bad&Bax, and DKK1, or activation of MMP-7, IL6, and Cyclin E/CDK2. These signaling pathways are involved in a wide range of biological activities such as cellular invasion, cellular migration, cell survival, and cellular proliferation, and are closely related to many cancers, making MSLN a notable immunotherapeutic target awaiting investigation.
