Hybridoma Rescue Strategies Overview

Hybridoma cells are highly valued for their ability to secrete antibodies and achieve unlimited proliferation. However, in practice, their activity can decrease or even become unusable due to improper selection or operation during the hybridoma cell process, as well as improper preservation after preparation. At this point, the existing hybridoma cells need to be rescued to restore their activity and ensure the smooth running of research. Creative Biolabs provides high-quality and efficient hybridoma rescue services, as well as custom antibody production based on hybridomas, to accelerate research.

Hybridoma Suspension Domestication

Hybridoma Rescue Strategies Overview

The composition of serum can be quite complex, containing various nutrients such as growth factors, hormones, and transfer proteins. The effects of these components on cells are not yet fully understood. Additionally, serum can differ from manufacturer to manufacturer and even from batch to batch, resulting in varying effects on cultured cells. This variation can lead to differences in culture conditions for different batches of hybridoma cells. To address this, hybridoma suspension domestication is performed by converting walled cells to suspension culture, which reduces the impact of unknown factors in the serum on hybridoma cell activity.

Hybridoma Re-cloning

Hybridoma recloning is a popular approach to rescuing highly active hybridoma cells. The process involves using methods such as limited dilution, semi-solid medium, soft agar, or automated systems. The specific method used for hybridoma recloning is chosen based on research needs and experimental conditions to ensure that the resulting hybridomas are highly active.

Hybridoma Enrichment

Enrichment of hybridoma cells with active cells by different methods is a powerful measure to restore the activity of hybridoma cells. In order to maintain the viability of hybridoma cells and their ability to produce antibodies, we need to enrich positive hybridoma cells to remove the rest of the cells and expand them. Commonly used methods for hybridoma enrichment are FACS-based enrichment and droplet-based microfluidics. In practice, the hybridoma cells can be enriched by selecting the appropriate method according to the specific research requirements.

Hybridoma Cell Line Optimization

To avoid reduced activity in hybridoma cells caused by contamination, inappropriate freezing methods, or prolonged freezing, we recommend performing hybridoma cell line optimization to identify the root cause before taking targeted action.

When dealing with hybridomas that have lower activity, it is essential to rule out contamination first. Mycoplasma contamination, viral contamination, and gummi contamination are common factors that can affect hybridoma cell activity and antibody production. Therefore, before taking any action, the cells should be bioassayed.

If the cells are less active due to freezing, the best approach is to establish a healthy culture of the cells by taking a scientific approach.

Lastly, if cell line drift is the issue, a comprehensive assay is generally required to remove abnormal cells and retain normal cells to ensure overall cell activity.

Hybridoma Re-fusion

Some hybridoma cells may exhibit reduced activity due to the loss of chromosomes from one of their parents. On the other hand, the re-fusion of hybridoma cells can potentially rescue their activity by reintroducing the corresponding parental chromosomal complement. Hybridoma re-fusion typically involves the fusion of hybridoma cells with myeloma cells. The key factor in this process is selecting the appropriate myeloma cells for fusion.
