Types of Anti-Idiotypic Antibody

There are three main types of anti-idiotypic antibodies based on their interaction with and detection of antibody drugs. The first type is antigen-blocking anti-idiotypic antibodies, which are characterized by having overlapping paratopes with specific sites of the antibody drug. As a result, the target antigen of the antibody-drug and the anti-idiotypic antibody compete with each other. Therefore, this type of anti-idiotypic antibody is capable of detecting only free antibody drugs. The second type of anti-idiotypic antibodies is known as non-containing antibodies. They are characterized by the absence of overlap between the paratopes and specific sites of the antibody drug. Consequently, both anti-idiotypic antibodies and antigens can bind to the antibody drug simultaneously without affecting each other's binding ability. Hence, non-contained anti-idiotypic antibodies are used to detect all forms of available antibody drugs, including both free and antigen-bound variants. The third class of anti-idiotypic antibodies is called complex specific anti-unique antibodies. This is because anti-idiotypic antibodies are unable to bind to an antibody drug unless the drug is already bound to its antigen. Therefore, these anti-idiotypic antibodies can exclusively detect bound antibody drugs.

Polyclonal anti-idiotypic antibodies are commonly used as a reference control for immunogenicity assays. Immunogenicity refers to the ability of a therapeutic agent, such as an antibody-drug, to induce a humoral and/or cell-mediated immune response, resulting in the production of anti-drug antibodies (ADA). These anti-drug antibodies negate the effects of the antibody-drug, essentially inhibiting its therapeutic aspects completely. Therefore, it is crucial for researchers to analyze the immunogenicity of new antibody drugs in preclinical analyses. Anti-drug antibodies are very similar to anti-idiotypic antibodies because both bind to the same antibody drug. Consequently, a pool of polyclonal anti-idiotypic antibodies can be used as a positive control when analyzing the presence of anti-drug antibodies in patient samples.

Since most anti-idiotypic antibodies are specifically produced against particular antibody drugs, they are commonly used in preclinical practice for pharmacokinetic analysis. Pharmacokinetics involves studying systemic drug metabolism, including the rate of absorption, distribution, bioavailability, and excretion of a drug in different patient cohorts. To achieve this, researchers need to track antibody drugs that are bound or unbound to their designated targets at different time points after administration. By using anti-idiotypic antibodies (primarily mAb), various forms of antibody therapy can be easily tracked and quantified in patient serum, blood, urine, or other body fluids.
