Description
2',7'-Dichlorofluorescein (DCF) is a classic fluorescent probe from the fluorescein family, established as a standard reagent for detecting reactive oxygen species (ROS) and quantifying intracellular oxidative stress in cell biology. The probe's core mechanism relies on an oxidation-activated, "turn-on" fluorescent response. Its common, cell-permeant, non-fluorescent precursor (e.g., H2DCF-DA) is hydrolyzed by intracellular esterases to the non-fluorescent 2',7'-dichlorodihydrofluorescein (DCFH). In the presence of various ROS, DCFH is rapidly oxidized to form the final DCF product, which exhibits intense green fluorescence (Ex/Em ≈ 504/529 nm). The increase in DCF fluorescence intensity correlates directly with cellular oxidative stress levels, making it a sensitive and reliable tool for studying redox homeostasis, assessing oxidative damage, and screening antioxidants.
