Description

EMCS is a non-cleavable, heterobifunctional crosslinker. It features an amine-reactive N-hydroxysuccinimide (NHS) ester and a sulfhydryl-reactive maleimide group, separated by a flexible 6-carbon (C6) aliphatic spacer. The NHS ester couples with primary amines (lysines) at pH 7.0-9.0. The maleimide group couples with thiols (cysteines) at pH 6.5-7.5. The long C6 spacer provides greater reach and flexibility than shorter linkers like BMPS or GMPS, helping to overcome steric hindrance when conjugating two large biomolecules, such as an antibody to an enzyme.
