Antibodies enable to bind almost any molecule with high affinity and specificity, offering potent biotechnological tools for diagnostic and therapeutic applications. Progresses in recombinant DNA technology have promoted the manipulation of the antibody genes, thus design, cloning, expression, and use of single-chain antibodies have become routine procedures in protein engineering. The potential of single-chain antibody fragments (scFv) for intracellular applications, named as “intrabodies,” has been studied in many laboratories. Intrabodies have been applied to target proteins in a singular fashion. Bispecific and tetravalent antibody fragments are able to promote and expand the inhibitory potential of intrabodies via displaying raised apparent affinity for their antigen and via being more efficient at inhibiting protein function or intracellular trafficking. Intrabodies stand for a powerful alternative to approaches of gene inactivation that target at the level of DNA or mRNA, for example, antisense, zinc finger proteins, targeted gene disruption, or the relatively new RNA interference.

A bispecific intrabody has two identical immunoglobulin chains paired via its recombinant Fc domains. Meanwhile, two pairs of single chain Fvs (scFvs) with distinct specificity are linked to each end of the Fc domain respectively. Since the component unit for dimerization is uniform, this strategy significantly simplifies the generation process of bispecific products. Furthermore, as each component unit is bispecific and bivalence, the homodimer product is not only bispecific but also tetravalence. These properties make bispecific intrabody a leading candidate format for both drug development and academic research.
A primary application for intrabodies lies in functional genomics. As the number of proteins with unknown function and their interactions with other proteins steadily rise, bispecific intrabody could offer an attractive proteomic tool to analyze these networks. Moreover, bispecific intrabody is used to study linked regulatory pathways or block redundant pathways. In addition, bispecific intrabody may extend the therapeutic applicability of intrabodies.
