Hepatocyte Growth Factor (HGF)

Hepatocyte growth factor (HGF) is a paracrine cellular growth, motility and morphogenic factor. It is secreted by mesenchymal cells and targets and acts primarily upon epithelial cells and endothelial cells, but also acts on hematopoietic progenitor cells and T cells. It has been shown to have a major role in embryonic organ development, specifically in myogenesis, in adult organ regeneration, and in wound healing. Hepatocyte growth factor regulates cell growth, cell motility, and morphogenesis by activating a tyrosine kinase signaling cascade after binding to the proto-oncogenic c-Met receptor. Hepatocyte growth factor is secreted by mesenchymal cells and acts as a multi-functional cytokine on cells of mainly epithelial origin. Its ability to stimulate mitogenesis, cell motility and matrix invasion give it a central role in angiogenesis, tumorigenesis, and tissue regeneration.

HGF Marker of Liver Failure

HGF exerts pleiotropic biological effects on different cell types. At least some of the biological effects of HGF are due to inhibition of apoptosis. HGF is a potent mitogen for hepatocytes and is crucial for liver development. Transgenic mice expressing HGF under the control of the albumin promoter performed liver regeneration two times faster than normal mice, whereas tumors of diverse origin were found in HGF transgenic mice under the control of the metallothionein promoter. Elevated HGF levels have been reported after partial liver resection, acute or chronic hepatitis, and fulminant liver failure. In acute hepatitis, serum HGF levels are approximately twofold higher compared with controls. Serial measurement of serum HGF levels in acute hepatic injury serves as a prognostic factor.
