Description
Biotin-HPDP is a thiol-reactive, cleavable biotinylation reagent. It features a 2-pyridyldithiol (HPDP) group that reacts specifically with free sulfhydryl groups (cysteines) to form a disulfide bond. This linkage is stable under normal conditions but is readily cleaved by reducing agents (e.g., DTT, TCEP). This reagent is used to biotinylate cysteine-containing proteins or peptides. Its key application is in affinity purification, where the biotinylated "bait" protein and its interacting partners can be captured on streptavidin and then gently released by cleaving the disulfide bond.
