Description
SMCC is a non-cleavable, heterobifunctional crosslinker widely used for creating stable Antibody-Drug Conjugates (ADCs). It contains an amine-reactive N-hydroxysuccinimide (NHS) ester and a sulfhydryl-reactive maleimide group. The two are separated by a rigid cyclohexyl spacer, which provides defined spacing and enhances *in vivo* stability. The NHS ester reacts with amines (lysines) on the antibody, and the maleimide reacts with thiols (cysteines) on the drug payload, creating a highly stable, non-cleavable thioether linkage.
