Description
CPO-PFP is a highly specialized crosslinker for the site-specific modification of N-terminal cysteine (N-Cys) residues. It features a highly reactive pentafluorophenyl (PFP) active ester and a cyclopropene (CPO) group. The PFP ester first acylates the alpha-amine of the N-terminus. This tethers the linker, positioning the CPO moiety for a rapid intramolecular reaction with the N-Cys thiol. This proximity-driven mechanism grants exceptional selectivity for N-terminal cysteines over internal cysteine residues, making it invaluable for creating homogenous, site-specific protein conjugates or for peptide stapling.
