Description
Synonyms: N-(9-Acridyl)maleimide(NAM), 1-(9-Acridinyl)-1H-pyrrole-2,5-dione. NAM is a thiol-reactive, "turn-on" fluorescent probe. The maleimide group reacts specifically with sulfhydryl groups (cysteines) at pH 6.5-7.5. The acridine-based reagent is essentially non-fluorescent in aqueous solution, but it becomes brightly blue-fluorescent upon covalent reaction with a thiol. This fluorogenic response makes it ideal for quantifying free thiols or for specifically labeling cysteine residues on proteins with minimal background fluorescence.
