Introduction of MMP-7

Matrix metalloproteinase-7 (MMP-7), also known as matrilysin, pump-1 protease (PUMP-1), or uterine metalloproteinase, is a secreted protease that belongs to the peptidase M10 family of matrix metalloproteinases (MMPs). MMP-7 plays a role in the breakdown of extracellular matrix in normal physiological processes, including reproduction, embryonic development, and tissue remodeling. Besides, it is involved in the pathogenesis of diseases such as arthritis and cancer metastasis. MMP-7 can break down proteoglycans, elastin, fibronectin, and casein. It lacks a conserved C-terminal hemopexin domain, which is the difference between MMP-7 and most of the other MMP family members. Studies have shown that MMP-7 is involved in wound healing due to its role in regulating the activity of defensins in the intestinal mucosa. It shows high expression levels in various human cancers.

MMP-7 Marker for Cholangiocarcinoma

Cholangiocarcinoma (CCA) is an aggressive and fatal cancer with a tendency for local invasion and distant metastases. However, the therapeutic methods for CCA have been limited since this type of cancer is poorly responsive to chemotherapy and radiation therapy. In order to improve the survival rate, patients suffered from CCA must be diagnosed and treated as early as possible. Previous studies have demonstrated that MMP-7 is found to be potentially involved in tumor metastasis and be frequently expressed in CCA specimens. High expression of MMP-7 facilitates cancer invasion and angiogenesis through degrading extracellular matrix macromolecules and connective tissues. The elevation of serum MMP-7 levels has already been proven as a useful marker for detecting cholangiocarcinoma, especially in those patients with obstructive jaundice.
