Description
TDBA is a heterobifunctional, photo-reactive crosslinker used for photoaffinity labeling (PAL). It possesses a carboxylic acid group, which can be activated (e.g., with EDC/NHS) to couple with primary amines on a 'bait' molecule. The other end features a trifluoromethyl-diazirine group. This moiety is stable in the dark but, upon activation with UV light (approx. 350-370 nm), generates a highly reactive carbene. This carbene non-selectively inserts into nearby C-H, N-H, or O-H bonds of an interacting 'prey' protein, permanently capturing transient interactions and enabling target identification.
