Description
DSS is a classic homobifunctional, amine-reactive crosslinker. It features two N-hydroxysuccinimide (NHS) ester groups separated by an 8-carbon (C8) aliphatic suberate spacer. NHS esters react efficiently with primary amines (lysines) at pH 7.0-9.0 to form stable, non-cleavable amide bonds. DSS is widely used to study protein-protein interactions by covalently "freezing" them. Its medium-length, hydrophobic spacer arm makes it membrane-permeable, allowing for crosslinking of proteins both in vitro and inside living cells. It is the non-cleavable analogue of DSP.
