Description
This heterobifunctional PEG linker features a thiol-reactive maleimide group and an acid-labile t-butyl (OtBu) ester, linked via a 4-unit PEG chain. The maleimide group is available for conjugation to sulfhydryl groups (cysteines). The t-butyl ester protects a terminal carboxylic acid. This protecting group is stable to the neutral-pH maleimide coupling but can be selectively removed using mild acid (e.g., TFA) to reveal the free carboxylic acid. This allows for a second, sequential coupling step (e.g., via EDC activation), making it useful for multi-step, orthogonal conjugation strategies.
