Introduction of Serial Passage Assay

The development and market launching of a new antibiotic always requires a comprehensive understanding of its possible risks, especially the emergence and spread of drug resistance in various bacterial pathogens. Resistance testing in the pharmaceutical industry can greatly reduce project termination and waste of resources due to resistance development during drug development. Numerous studies showed that antibiotic resistance mechanisms always impair bacterial fitness, such as the reduction of growth rate. In this case, serial passage assay has been the most used method to predict the emergence of resistance to novel antibiotics. What’s more, it is also useful to detect genes in which mutation can confer resistance, as well as to analyze compensatory mutations. However, please note that it is not able to address the risk of resistance development due to horizontal gene transfer (HGT).

Procedure for Serial Passage Assay

This assay involves the serial passage of interesting bacterial strains in media containing antibiotic or AMPs at concentrations near the MIC, which would be increased with the enrichment of resistant mutants. After different time points, the bacterial are recovered for broth-dilution MIC test and time-kill assay to determinate the resistance. It is relatively easy to perform and the small growth volumes require low amounts of samples. Studies have shown that following only four serial passages, Staphylococcus aureus resistance to AMP lactoferrin B increased more than 30-fold.
