Baculovirus Vector


Viral vectors have been utilized in gene therapy to delivery therapeutic genes to target cells through ex vivo or in vivo methods. The baculovirus is an insect virus that can efficiently transduce mammalian cells for transgene expression. It has emerged as a novel vector for gene delivery Because it is non-pathogenic to humans and causes minimal toxicity to mammalian cells. Baculoviruses are enveloped viruses with a circular dsDNA genome. As the model species of baculoviruses, the autographa californica multiple nucleopolyhedrovirus (AcMNPV) in a mammalian expression cassette is highly effective in mediating transient transgene expression in human cells. The baculovirus vector genome is approximately 130 kb with a large packaging capacity (allowing at least 38 kb of inserts), especially suited for short-term and high-level transgene expression.

As vaccine carriers, baculovirus vectors can simultaneously stimulate the humoral and cellular immune responses for the treatment of influenza, malaria and rabies. Baculoviruses can be transduced at extremely high efficiency in primary chondrocytes, bone marrow–derived mesenchymal stem cells (BMSCs), and adipose-derived stem cells (ASCs). By expressing various growth factors, baculovirus gene therapy can be used in tissue engineering and regenerative medicine, for example, to regulate cell differentiation and improve cartilage formation.

Baculoviruses also can express suicide genes, tumor suppressors or other antitumor genes at high efficiency in specific cancer cells. And we provide the design of inserts with immunepotentiating genes to elicit strong host immunity against malignancies of the bladder, brain, lungs and prostate.

Manufacturing Process of Baculovirus Vector
