Background of Hybridoma Technology

Hybridoma is a technique for animal cell fusion. In 1975, two British scientists found that the fused cells had the characteristics of parental cells and could be developed into simple cell lines by cloning.

According to this principle, a group of hybridoma cells that can infinitely produce antibodies can be obtained by fusing antigenic animal spleen cells (commonly used mouse B lymphocytes) with tumor cells that can reproduce infinitely (commonly used mouse myeloma cells). After single cloning, the cell population can continuously secrete a single specific monoclonal antibody (mAb). At present, the production of monoclonal antibodies using hybridoma technology has become one of the most valuable achievements in animal cell fusion.

Hybridoma Limitation & Solution

Although hybridoma cells have great advantages in antibody production, this technique still has some shortcomings.

(1) Antibody quality fluctuation: antibody production depends on whether mice can produce target-specific antibodies, and the effect is affected by individual health status, immune status, and other factors. The consistency and repeatability of antibodies produced by different hybridoma cells are poor.

(2) Allergic reaction: this technique usually uses mouse cells for monoclonal antibody production, and animal-derived antibodies may cause allergic reactions in patients, limiting the use of hybridoma technology as clinical therapy.

(3) Loss of antibody specificity: although hybridoma cells can proliferate indefinitely and express antibodies, long-term cell proliferation and protein expression may lead to antibody gene mutation and invalidation of expressed antibodies.

(4) Cryopreservation loss: hybridoma cells usually need to be cryopreserved, and the cryopreservation process will also cause the disappearance of their monoclonal antibody specificity.

An effective method to solve the heterogeneity of hybridoma cell antibody quality and the limitations of sensitization and specificity loss is hybridoma cell antibody sequencing.

What Is Hybridoma Sequencing?

The sequencing of hybridoma cells is to reverse transcribe the RNA extracted from hybridoma cells to obtain cDNA, then amplify it with PCR, and then obtain the complete sequence information of the heavy and light chains of the antibody. Compared with monoclonal antibody amino acid sequencing, gene sequencing can more accurately identify amino acids that are difficult to distinguish in mass spectrometry identification, so it is more accurate.

The sequenced monoclonal antibodies will be preserved, transmitted, communicated, and identified in the form of base sequences. When needed, stable and large-scale antibody production is carried out by recombinant protein expression technology according to the sequence. In the follow-up process of antibody production, hybridoma cell culture and cell cryopreservation are no longer needed to avoid the loss of antibody specificity. In addition, the production of recombinant protein does not involve the process of immune response, which avoids human allergic reactions caused by animal-derived antibodies. The antibody can also be modified during the construction of the recombinant vector, such as antibody fragment expression, homotype replacement, chimeric antibody, and antibody humanization. In addition, the monoclonal antibody expressed by the recombinant protein has good consistency, high repeatability, and little quality difference between batches.
