Protein isoform formation

Proteins are structurally heterogeneous molecules and the same protein can exist in many isoforms (sub-populations). A set of protein isoforms may be present and formed due to alternative splicing, polymorphism and post-translation modifications (PTMs). For examples, actin isoforms are originated from separate genes, because single gene can code for multiple proteins during the mRNA splicing. In addition, a glycoform is an isoform of a protein that differs only with the number or type of attached glycan. Therefore, glycoproteins often have a lot of glycoforms which may result from variation in the carbohydrate structures during the process of glycosylation.

Importance of protein isoform

Protein isoforms are involved in various stages in a number of pathologies, playing important roles in various biological processes. Therefore, they have potentially been used as biomarkers and therapeutic targets. For example, several specific proteins isoforms are related to thyroid pathological conditions and various cancers, multiple sclerosis and autoimmune diseases. Therefore, it is very important to detect the protein isoforms.

Protein isoforms detection method: MAIIA

The existing separation and detection methods for protein isoforms, such as chromatographic separation and electrophoretic separation, are far too complicated and time-consuming.

Based on the extensive experience in protein engineering, scientists of Creative Biolabs have built up an innovative membrane-assisted isoform immunoassay (MAIIA) method for protein isoform detection. This technique combines the separation of protein isoforms (by ion-exchange or affinity chromatography) with an immunoassay detection method in a disposable membrane device. It is possible to quantify the protein isoforms in a very fast and simple way (<15 min). In addition, it has achieved much higher levels of specificity than the traditional methods. This technique has been successfully applied in measuring carbohydrate-deficient isoforms of the glycoprotein transferrin. Notably, with our MAIIA technology, we are able to resolve and detect other types of post-translational modified proteins even when they occur in low concentration. Our technique has been widely used for rapid testing of clinically interesting protein isoforms.
