Interactions between cancer cells, immune cells, and fibroblasts result in changes within the tumor microenvironment (TME), which further contributes to increased tumor heterogeneity. TME exerts pivotal influences on malignancy initiation, tumor progression, and metastasis. Traditional 2D models derived from patient blood or tumor samples fail to robustly preserve the intricate diversity and physical structure of the TME. By co-culturing tumor organoids derived from patients with adaptive immune cells, innate bone marrow cells, cancer-associated fibroblasts, lymphocytes, et al., we have constructed a complex and dynamic tumor organoid-TME co-culture model that facilitates spatiotemporal interactions, enabling systematic analysis of key factors in tumor recognition or anti-tumor immune responses.
Through co-culturing tumor cells and TME cells from patient sources, our tumor organoid spheres serve as dynamic, highly regulated, and evolving physiological models.
Co-culturing different types of tumor cells with specific TME cells can provide a wider range of models and effectively predict the efficacy of immunotherapy or drugs.
With various combinations of tumor cell types and TME cells, our model is applicable to drug screening, genomic research, radiotherapy screening, or cancer gene identification.
