TILs (tumor-infiltrating lymphocytes) exhibit specific anti-tumor immune activity and are populations of lymphocytes that penetrate deep inside tumors for immune assault. Initial cell distribution and multiple recombinational analyses in co-culture models were investigated. The study of time-dependent migration patterns of TILs permits simultaneous longitudinal detection of TIL activation and cytokine secretion, which can be used as a key indicator for assessing adaptive responses.
Through the co-culture model, the dynamic killing effect of TILs on tumor cells can be assessed in real-time, advancing the establishment of an anti-tumor overt TIL strategy.
To study tumor immune escape pathways involving TILs, including immune checkpoint regulation and cellular depletion, and to develop personalized strategies to overcome tolerance.
To simulate the effect of drugs in vivo on the clustered tumor environment of TILs in actual lesions, the degree of infiltration and distribution of TILs in 3D organoid co-culture models were used as predictive indicators to screen effective drugs for targeted modulation.
Co-culture models of tumor organoids and autologous or non-autologous immune cells help to overcome the limited information available in traditional models.
