Description
This is a key building block for Antibody-Drug Conjugates (ADCs). It features an Alloc-protected (allyloxycarbonyl) amine, a cathepsin B-cleavable Valine-Citrulline (Val-Cit) dipeptide, and a p-aminobenzyl alcohol (PAB-OH) self-immolative spacer. The Alloc group is an orthogonal protecting group that can be removed with Pd(0). This linker is an intermediate used to construct protease-cleavable ADCs, where the PAB-OH end is typically attached to a drug payload before conjugation to an antibody.
