Description
DSP, also known as Lomant's Reagent, is a homobifunctional, amine-reactive, and cleavable crosslinker. It features two N-hydroxysuccinimide (NHS) ester groups that react with primary amines (lysines) at pH 7.0-9.0. The key feature is that the 12-angstrom spacer arm contains a central disulfide bond. This bond is stable during conjugation but can be easily cleaved using reducing agents like DTT or TCEP. This cleavability makes DSP ideal for two-stage applications, such as identifying protein-protein interactions (e.g., separating crosslinked complexes on a 2D-PAGE gel).
