Description
This is a heterobifunctional, reducible PEGylation crosslinker. It contains an amine-reactive NHS ester and a thiol-reactive pyridyldithiol (SPDP) group, connected by a 4-unit PEG spacer. The NHS ester reacts with amines. The SPDP group reacts with thiols to form a disulfide bond. This bond is readily cleaved by reducing agents (e.g., DTT, or intracellular glutathione). This cleavability is essential for its use in drug delivery, particularly as a linker for Antibody-Drug Conjugates (ADCs), where release of the drug payload inside the target cell is required. The PEG4 spacer adds hydrophilicity.
