Introduction of DNA Restriction Enzyme Spectrum Analysis

Restriction enzymes, also known as restriction endonucleases, are DNA-cutting enzymes isolated from bacteria. These enzymes are thought to play an important role in host defense by cutting and destroying viruses or other foreign DNA. Restriction endonuclease recognizes specific palindromic DNA sequences and cuts the double-stranded DNA into fragments at specific recognition sites. These sites are called restriction sites, normally ranging from 4-8 base pairs in length. Now there are many different types of restriction enzymes that have been identified such as EcoRI, BamHI, and HindIII. Each type recognizes a different restriction site. For example, BamH1 specially recognizes the sequence of GGATCC and EcoRI specially recognizes the sequence of GGATCC. Restriction enzymes have a variety of applications in biological fields such as molecular cloning, single-nucleotide polymorphisms (SNPs) identification, and DNA fingerprinting. Furthermore, DNA restriction enzyme analysis is increasingly used as a tool for the diagnosis of some genetic disorders.

Application of Mstll Restriction Enzyme in IVD for Sickle-cell Anemia

Sickle cell anemia is a genetic disease of the red blood cells (RBCs) that is caused by a single-base mutation of the β-globin gene. In normal people, the β-globin gene sequence is CCTGAGGAG, while in sickle cell anemia, the β-globin gene sequence is CCTGTGGAG. The single-base mutation (A to T) in the β-globin gene destroys the recognition site of the Mstll. Therefore, the mutation can be detected by using restriction enzyme Mstll to cut DNA fragments in and around β-globin gene, followed by performing a hybridization with a cloned β-globin DNA probe.
