Overview of BPTF

In humans, the nucleosome-remodeling factor subunit (BPTF, also known as bromodomain PHD finger transcription factor, fetal Alz-50 reactive clone 1 or FAC1) is a protein encoded by the BPTF gene located at chromosome 17q24.3. BPTF is identified in patients with Alzheimer's disease for the first time and upregulation of BPTF is found in patients with neurodegenerative diseases. BPTF plays an important role in transcriptional regulation because of its DNA-binding domain and zinc finger motif. BPTF is important to embryonic development and is associated with ATP-dependent chromatin remodeling.

BPTF in Disease

Associated diseases of BPTF contain neurodevelopmental disorders and cancers such as melanoma and hepatocellular carcinoma. It is presumed to be an oncogenic element because of its existing in a series of tumors.

  • Melanoma

BPTF functions as an oncogene in the progression of melanoma development. Upregulation of BPTF was observed and predicted for poor survival in patients with melanoma. When BPTF is knockdown in an shRNA-mediated pathway, the proliferative capacity and metastatic potential of melanoma cells are inhibited. Overexpression of BPTF enhances the drug-resistant of the melanoma cell line to the BRAF inhibitors. BPTF has protumorigenic functions and can be identified as a novel target for anticancer therapy.

  • Hepatocellular carcinoma

High expression of BPTF means poor survival in patients with hepatocellular carcinoma (HCC). The expression of BPTF is associated with vascular invasion, tumor number, recurrence, and so on. Upregulation of BPTF and vimentin and decreased expression of E-cadherin are found in HCC, and high expression of BPTF is significantly related to the elevated expression of vimentin and decreased E-cadherin by Spearman rank correlation analysis, which means BPTF is a critical regulator of epithelial-mesenchymal transition in HCC. BPTF can independently predict the overall survival and disease-free survival of patients after hepatectomy.
