Description
SIA is a classic heterobifunctional, non-cleavable crosslinker. It features an amine-reactive N-hydroxysuccinimide (NHS) ester and a sulfhydryl-reactive iodoacetyl group. The NHS ester reacts efficiently with primary amines (lysines) at pH 7.0-9.0. The iodoacetyl group is an alkylating agent that reacts specifically with thiols (cysteines) at pH 7.5-8.5 to form a highly stable, irreversible thioether bond. While it reacts slower than a maleimide, the iodoacetyl group is less prone to hydrolysis. It is used for short-range, stable crosslinking of amines to thiols.
