Description
Synonyms: N-Succinimidyl 3-(2-pyridyldithio)propionate. SPDP is a classic heterobifunctional, cleavable crosslinker. It features an amine-reactive N-hydroxysuccinimide (NHS) ester and a sulfhydryl-reactive 2-pyridyldithiol group. The NHS ester couples to primary amines. The 2-pyridyldithiol group reacts with a free thiol (cysteine) to form a new disulfide bond. This reaction releases pyridine-2-thione (monitorable at 343 nm). The resulting disulfide bond is readily cleaved by reducing agents (e.g., DTT, TCEP), making this linker ideal for reversible crosslinking, affinity purification, or creating reducible bioconjugates.
