Overview of NCBP1

In humans, nuclear cap binding protein subunit 1 (NCBP1, also known as NCBP 80 KDa Subunit) is a protein encoded by NCBP1. NCBP1 is a component of the nuclear cap-binding protein complex (CBC), which is an RNA-binding protein. CBC involves a series of cellular processes such as pre-mRNA splicing, nonsense-mediated mRNA decay, and translational regulation by binding to the monomethylated 5' cap of nascent pre-mRNA in the nucleoplasm. Besides, CBC containing NCBP1 can also promote 3'-end processing and RNA nuclear export.

NCBP1 in Disease

Ectopic expression of NCBP1 is associated with cassava brown streak disease in plants and cancers such as lung cancer and breast cancer.

  • Lung cancer

NCBP1-CUL4B oncoprotein axis is linked to lung cancer progression. Lung cancer is one of the most frequent cancer types with a high correlation with tumor-associated deaths worldwide. It is reported that silencing of NCBP1 leads to cell growth reduction of HeLa cells. Compared with non-lung cancer tissues, upregulated expression of NCBP1 can be identified in lung cancer tissues, together with several lung cancer cell lines. In lung cancer cells, overexpression of NCBP1 promotes cell growth, wound healing, migration, and epithelial-mesenchymal transition, while knockdown of NCBP1 can reverse it. CUL4B, as a downstream target gene of NCBP1, can be upregulated by NCBP1 in NSCLC. Silence of CUL4B will reverse the tumorigenesis of lung cancer cells caused by NCBP1. Taken together, NCBP1-CUL4B sheds new light on the treatment of lung cancer.

  • Breast cancer

Elevated expression of NCBP1 is related to the basal subtype of breast cancer (BC). Classification of disease subtypes will enhance diagnostic capabilities and the acquisition of prognostic information. The subtypes of human BC include basal or basal-like, luminal A, luminal B, and HER2+. According to the microarray data, transcriptome-wide high expression of NCBP1 can be observed in basal-like BC patients. Upregulated NCBP1 expression predicts better overall survival. Thus, NCBP1 primary tumor expression can distinguish the basal subtype of BC from other subtypes and can be a positive prognostic indicator in basal BC patients.
