Advantages of non-human primates (NHPs) neurological models

Non-human primates (NHPs) have a vertebrate brain with similar neural circuitry to humans. Moreover, they also have such similarities with human beings in physiological and behavioral characteristics. All these features make them more accurate and valuable models of neurological diseases than other animals, constituting an indispensable tool for many PK/PD studies.

What is Parkinson's disease (PD)?

Parkinson's disease (PD) is a debilitating neurodegenerative disease. The onset of Parkinson's disease is insidious and progression is slow. The initial symptoms are usually shaking, rigidity, slow movement, and walking difficulty while thinking and behavioral problems may also appear. Non-motor symptoms such as depression, constipation, and sleep disorders are also common complaints in patients with PD. They affect the life quality of patients even more than those of motor symptoms.

What can we contribute to the investigation of Parkinson's disease (PD)?

In the past decades, NHP models of PD have been established in a variety of formats such as aging, genetic mutations (e.g. α-synuclein, parkin, LRKK2, PINK1, DJ-1) and neurotoxins (6-OHDA, MPTP, pesticides, rotenone, reserpine). Creative Biolabs provides two widely used PD models:

  • 6-OHDA and MPTP induced PD models. 6-OHDA cannot cross the blood-brain barrier and requires the intracerebral administration to exert its toxic effects via oxidative stress. But this model may provide a window of time to study oxidative stress and neurodegeneration and to test neuroprotective strategies by inducing a retrograde, relatively progressive nigrostriatal lesion.
  • The other model, MPTP, is the most widely used neurotoxin for building PD model. MPTP is highly lipophilic and able to cross the blood-brain barrier. MPTP-treated monkeys replicate representative PD features such as rigidity, bradykinesia, and postural instability while nonmotor signs such as frontostriatal cognitive deficits and sleep pattern changes may also appear. In Creative Biolabs, we have rich experience in constructing both unilateral and bilateral MPTP-induced Parkinson's disease models.

Monkey models of PD can present behavioral and neurochemical deficits that are characteristic of humans PD patients. Behavioral tests provide an external measure of the brain and have been a primary outcome measure when assessing treatments in animals. We provide a full set of behavioral tests using a rating scale, fine motor skill tasks, and activity monitoring, etc. Besides that, general measures such as regular body weight and temperature records, standard blood and chemistry panels, frequency and characteristics of feces are also included. Moreover, we provide in vivo functional imaging with positron emission tomography (PET) or magnetic resonance imaging (MRI) to assess neuronal connectivity.
