Membrane proteins act as one critical class of medical targets against various disease types. Nowadays, antibodies targeting membrane proteins have become attracting increasing research interest, due to the high specificity, high affinity, low cytotoxicity and versatile functions. However, it can be quite challenging to raise ideal binders against membrane proteins. For one, membrane proteins are generally large molecules with multiple domains, which is very difficult to isolate and purify; besides, binders shall be able to elicit proper cellular functionalities. Therefore, a prerequisite is to have large pool of binders first. Traditional hybridoma is restricted by limited cell fusion efficiency that can lead to loss of many promising candidates. Phage display, on the other hand, is able to create a massive library with large capacity, but the library members have lost original VH-VL pairs, which can be problematic in downstream development.
