As a unique mammal species, camels brings a brand-new idea into antibody development. It has a novel antibody isotype termed as heavy-chain only antibody, which is merely composed of two identical heavy chains without light chains. The heavy-chain only antibodies contain a single monomeric variable region in Fab domains (aka VHH fragments) and count for about 50% of the antibodies in camelids. VHH fragments can be further developed into single domain antibodies (sdAbs), which represent the smallest antibody format with diagnostic and therapeutic activities.
VHH antibodies have many appealing features. The CDR3 loop of VHH domain is much longer than that of conventional antibodies and sometimes stabilized by a disulfide bond, which contributes to binding in cavities, such as enzyme and receptor clefts. Moreover, thanks to their small size, sdAbs are more robust, flexible but with similar specificity and affinity as conventional antibodies. It’s also much easier to transform sdAbs into economical bacterial cells easier, which is more ideal for large-scale production.
Anti-membrane protein antibodies are more and more appealing in clinical trials and therapeutic research.
