Introduction of Minimum Inhibitory Concentration (MIC)

Minimum Inhibitory Concentration (MIC) is defined as the lowest concentration of an antimicrobial ingredient or agent that inhibits the visible growth of microorganisms after overnight incubation. MIC has been served as the standard research tool for the determination of the in vitro activity for new antimicrobials. In general, broth dilution assay and Etests are two classical methods for MIC determination. During the development of antibiotics, MIC assessment is also essential to determine the appropriate concentration required for the final product and to ensure effective selection of antibiotics to increase the success rate of treatment. Correspondingly, the minimum bactericidal concentration (MBC) is the lowest concentration of antimicrobials that causes bacterial death. In general, the closer the MIC is to MBC, the stronger the bactericidal activity of the antimicrobials.

Standard CLSI M07-A9 for MIC Test

Established by the Clinical and Laboratory Standards Institute (CLSI), CLSI M07-A9 is always the gold standard for MIC determination. To perform this test, serial dilutions of test antimicrobials are made in a liquid microbial growth medium, while the medium is inoculated with standardized numbers of microorganisms individually for a defined period of time. At this point, the lowest concentration (highest dilution) of test antimicrobials that prevents turbidity is the MIC.

Because viable bacterial cells can reduce the non-fluorescent resazurin (blue) to the fluorescent resorufin (pink), resazurin can also be used to provide direct quantitative measurements of bacterial metabolic activity and viability. To ensure the accuracy and reproducibility of experimental results, each sample will be tested in triplicate.
