Produced by bacteria, β-lactamases are enzymes present multi-resistance to β-lactam antibiotics, including penicillins, cephalosporins, cephamycins, monobactams, as well as carbapenems. These antibiotics have a common element in their molecular structure, the β-lactam ring. β-Lactamase can open the β-lactam ring through hydrolysis, thereby destroying the structure of antibiotics and inactivating the antibacterial properties. The β-lactamases can be divided into two forms, including Streptomyces serine β-lactamase (SBLs) and Metallo-beta-lactamases (MBLs). For example, penicillinase is the first defined β-lactamase that showed specificity for penicillins by hydrolyzing the β-lactam ring.

Creative Biolabs provides a simple method to measure β-lactamase activity in food, fermentation samples, and biological samples. One unit of β-lactamase can be defined as the amount of enzyme required to hydrolyze 1.0 μmole of nitrocefin per minute at pH 7.0 and 25 °C.
The β-lactamase activity is measured by hydrolyzing nitrocefin, which is a chromogenic cephalosporin substrate that has been widely used for the detection of β-lactamase enzymes produced by various microbes. Ultimately, a colorimetric product (A490) can be obtained that is proportional to the enzymatic activity present. What's more, this assay can be performed in a 96-well or 384-well plate format for throughput screening of inhibitors against β-lactamases.
