The bacterial protease units can be defined as the quantity of enzyme that produces the equivalent of 1.5 μg/mL of L-tyrosine per minute at pH 7.0 and 37°C. The assay is based on the proteolysis of casein, where unhydrolyzed casein is removed by filtration and the dissolved casein is measured spectrophotometrically. In this assay, the substrate blank absorbance would be served as a reference to reflect the integrity of the assay and evaluate the reagent variability. After three replicates, the difference in results should be limited to 3%.
