Ang2 (Angiopoietin-2), also referred to Ang-2 or ANGPT2, is a 57 kDa glycoprotein encoded in humans by the ANGPT2 gene. It is located at the chromosome of 8p23.1 and belongs to the family of vascular growth factors. Ang2 is established to regulate vascular remodeling by facilitating endothelial cell survival, proliferation, and migration, as well as destabilizing the interaction of EC and perivascular cells. There are Ang1 (Angiopoietin-1) and the orthologs 4 also included in this family. The endothelial TEK tyrosine kinase Tie1 and Tie2 are receptors for angiopoietins. Ang2 binding to the Tie2 works as an antagonist without activating this receptor. It is also an antagonistic factor of Ang1 by disrupting the vascular remodeling ability of Ang1 and likely inducing endothelial cell apoptosis. VEGFA (Vascular endothelial growth factor A), also known as VPF, VEGF or MVCD1, is the most potent growth factor for endothelial cells and belongs to the PDGF/VEGF growth factor family. In humans, it is encoded by the VEGFA gene localizing at the chromosome 6p21.1. VEGF-A, as well as VEGF-C, and -D are found in the lung. During the embryonic and pseudoglandular stages, it is expressed in epithelial and mesenchymal compartments, then becoming more restricted to the epithelium as development process. VEGFA encodes a heparin-binding protein existing as a disulfide-linked homodimer. This growth factor is involved in inducing proliferation and migration of vascular endothelial cells. It is essential for physiological and pathological angiogenesis and considered as a powerful vascular permeability-enhancing agent.
