Introduction of Excretion Balance Study
As an important part of in vivo ADME studies, excretion balance studies are conducted to investigate the percentage of the dose eliminated. What’s more, urinary, biliary and fecal excretions are relative importance for data analysis. Generally, the study is performed in general toxicity species (both rodent and non-rodent), later in development also in the second carcinogenicity species. With animals kept in metabolic cages, excreta are carefully collected, volume or weight recorded and total radioactivity concentration in each sample measured at various time intervals up to generally 5 or 6 days, or longer if the half-life of radioactivity is long and excretion of radioactivity is still notable. Furthermore, some samples, the ones where radioactivity is more concentrated, are appropriately treated and analyzed with chromatographic, spectrometric and radiometric systems to identify and quantify the main metabolites.
