Quantitative Tissue Distribution Studies

Tissue distribution study plays a vital role in drug development programs as it can provide important information on the distribution of test compound into tissues of the animals selected for preclinical assessment. Theoretically, each part of the body can receive different doses of the drug and remain the drug in the different amount of time. Traditionally, investigations are performed by conventional methods, such as tissue dissection, total radioactivity determination, and LC-MS-MS/QPCR. However, the appearance of quantitative whole-body autoradiography (QWBA) has reformed the drug distribution research by utilizing phosphor imaging technology and offering high-resolution images.

Quantitative Whole-body Autoradiography (QWBA)

Quantitative whole-body autoradiography (QWBA) is a widely used method utilizing phosphor imaging technology to determine the distribution and concentrations of radiolabeled test compounds in laboratory animals. The most significant advantage of QWBA is that drug concentrations reflected by this technique can be maintained at the time point of euthanasia which cannot be obtained by any other techniques at present. Importantly, QWBA can provide information on tissue PKs, penetration, accumulation and retention.
