Background

IL-1α and IL-1β, both vital constituents of the Interleukin-1(IL-1) family, are potent cellular activating cytokines, despite being encoded by distinct genes and exhibiting relatively low sequence homology.

IL-1α is widely recognized as an endogenous danger signal, ubiquitously residing within the nucleus of many cells, and explicitly released during necrosis. IL-1β, on the other hand, is a potent pro-inflammatory cytokine and a pivotal mediator of inflammatory responses, indispensable for the host's defensive reactions to infections and injuries.

IL-1α and IL-1β bind with discrepant affinities to the IL-1R1 located on responsive cells, consequently recruiting accessory proteins, which associate with the MyD88 adapter molecule. This alliance triggers a cascade of downstream signaling events, leading to the activation of p38 and JNK kinase signaling and the subsequent release of the transcription factor NF-κB. Both IL-1α and IL-1β are discerned as equivalently efficient inflammatory cytokines. Consequently, dysregulation within their signal transduction pathways can underpin the onset of severe chronic or acute inflammatory diseases.
