Live animal-based SELEX Services for Aptamer Development

Introduction of Live-animal-based-SELEX

Live-animal-based-SELEX, also known as in vivo-SELEX, is developed to overcome some problems associated with in vitro protein-SELEX and cell-SELEX. Aptamers selected by protein-SELEX or cell-SELEX have shown low toxicity and immunogenicity, which greatly hinder the application of the aptamers in disease diagnosis and therapy. In vivo-SELEX is applied to select functional aptamers targeting organs or tissues of a living animal, which greatly improves the affinity and specificity of aptamers in vivo. The in vivo-SELEX starts with the construction of a random library of RNA or DNA aptamers modified by 2´-fluoropyrimidine and then systemically injects the library into a living animal. Certain aptamers bind to the desired target organs or tissues and unbound aptamers are eliminated by the kidney. Whereafter, the target organs or tissues are harvested for the extraction of bound aptamers. The extracted aptamers are amplified by PCR for the next round of screening. After 15~20 rounds of screening, the aptamers specific to target organs or tissues are enriched and then identified by high-throughput sequencing. In vivo-SELEX has been employed to develop DNA/RNA aptamers targeting brain tissues and certain tumor tissues such as intrahepatic colorectal cancer and lymphoma.
