What is ELONA?

Enzyme-linked immunosorbent assay (ELISA) is a fast and easy-to-use method for target detection through immunologic reactions and enzyme immunoassay, which has been commonly applied for preclinical and clinical quantification for several decades. Recently, a novel and improved ELISA, named enzyme-linked oligonucleotide assay (ELONA), has drawn our attention, showing high accuracy and low batch differences with relatively lower cost.

Similarly, ELONA relies on aptamers, a class of single-stranded oligonucleotides with certain spatial structures that can recognize targets with high specificity and affinity, to detect the target based on the principle of ELISA. ELONA generally comprises several variations according to the difference of aptamer or antibody/aptamer.

A number of aptamers have been developed through in vitro systematic evolution of ligands by exponential enrichment (SELEX) owing to their excellent performances, such as stable structure, high affinity (pM-nM), high specificity, and lower batch-to-batch variations. Our ELONA Development Process

Screening & selection

Screen and select the candidate aptamer with high specificity and proper modifications. The lead time will be shorter if your desired aptamer is ready to use on our web. And, to be more convinced, a special antibody used as a control will be included. Also, the appropriate target concentration should be confirmed.

Matrix compatibility

When the aptamer is ready, sample preparation condition evaluation and exploration of linearity or matrix impact on blank measurement are needed to ensure the accuracy of detection and eliminate some unnecessary interference factors.

Process optimization

An ELONA is beginning to take shape after a round of screening and exploration. What we need to do next is to optimize the conditional and process, such as control settings, calibrator concentrations, incubation time, incubation temperature, aptamer concentrations, and modifications.

Assay optimization and robustness

A seasoned ELONA will be generated after assessments on repeatability, multiplexing, cross-reactivity, formulation, design specification, SOP applicability, and some others.

Step 1: Screening & selection.
Aptamer candidates were obtained by in vitro SELEX services. Immunoassays are performed to compare the performance of different aptamer candidates (Clickmer 1, 2, 3 in Fig. 8) with three concentrations of a target protein.

Step 2: Matrix compatibility tests.
Test the impact of different matrix concentrations on the assay signal by sandwich immunoassays. Fig. 9 shows the detection effects of target protein in different matrices, including no serum, 12.5% serum, 25% serum, 50% serum, and 50% serum no target.
