Background of In Vitro Analysis for Aptamer

Nucleic acid aptamers are single-stranded DNA or RNA oligonucleotide sequences that bind to specific target molecules with high affinity and specificity through the ability to adopt a three-dimensional structure in solution. Aptamers have great potential as targeted therapeutic agents, diagnostic agents, delivery agents and biosensors. However, aptamers composed of natural nucleotide monomers degrade rapidly in vivo and show poor pharmacodynamic properties. To overcome this, many chemical modification techniques have been used to optimize the stability and toxicity of aptamers. Therefore, whether it is a natural aptamer obtained through SELEX (Systematic Evolution of Ligands by Exponential Enrichment) screening or a chemically modified aptamer, in vitro analysis is an important part of evaluating the success of aptamer development.
