Recombinant human C3a and its desArg variant were produced in Escherichia coli and purified using a sequential chromatographic protocol. Initially, cells were subjected to Ni2±NTA affinity purification in two stages, ensuring the capture of the target proteins, followed by an ion-exchange step that further refined purity. On the cation-exchange column, C3a and C3a desArg emerged as single, monodisperse peaks, eluting at 380 mM and 330 mM NaCl respectively, reflecting their distinct isoelectric properties. The overall yield was approximately 0.5–0.7 mg per liter of bacterial culture. These data confirm protocol consistency.
