Anaphylatoxins, also known as complement peptides, are fragments produced as parts of the activation of the complement system. Among them, C4a has the lowest activity. Through years of hard work and experience, Creative Biolabs has established a series of techniques and protocols for better research of the complement system. Here, we describe the anaphylatoxin C4a purification protocol for our clients all over the world.

Serum protein analysis in IgA nephropathy revealed that a specific signal persisted after control antibody immunoprecipitation yet was ablated using anti-C4a, suggesting it corresponds to C4a or its desArg variant. Its identity was verified by Western Blotting. Moreover, dilution of purified C4a desArg in sodium acetate (pH 4.5) yielded a concentration-dependent signal on surface-enhanced laser desorption/ionization (SELDI) analysis, with spiking into control sera reproducing the patient profile. Notably, patients exhibited significantly elevated serum C4a desArg levels, which correlated with glomerular lesion severity and mesangial hypercellularity. These findings indicate that serum C4a desArg may serve as a reliable biomarker for IgA nephropathy severity.
