The hemolytic potential of various C3 preparations was evaluated using a classical pathway hemolysis assay. The lysis of 0.05% IgM sensitized erythrocytes, i.e., the hemolytic potential of C3 was evaluated by incubating cells in C3-depleted serum supplemented with native C3, C3b, pH 11-treated C3, or various freeze/thaw C3 preparations. Specifically, one preparation comprised precipitated and native C3 subjected to five cycles between –20°C and room temperature, while another involved C3 cycled five times from –80°C to 37°C. Complete lysis was achieved by exposing cells to pure water, and the extent of hemolysis was quantified by measuring absorbance at 414 nm. These findings influence investigations into the structural configuration and functional properties of C3.
